I have a couple "sing-along" blu-rays that I want to rip and have them work like they do on the discs.
In one case (Mamma Mia), if you pick the last English subtitle is shows up with just the words of the songs and they're in a colored box with highlighting as each word is sung.
Is there a way to make that happen, or is that a disc-only thing?
I've been using MAKEMKV and HANDBRAKE to create MKVs for my Plex server.
Try StreamFab Downloader and download from Netflix, Amazon, Youtube! Or Try DVDFab and copy Blu-rays!

What type of subs are they? If they are srt then you won't get any of those fx's. To use with a mkv file and depending on your playback device you need .ass format. There was a program called PunkGraphicStream ASS to Blu-ray subtitle converter ie ASS to SUP for changing an ASS sub with all the fx's into the image format that blu-ray uses. So I guess you need the reverse. Convert SUP to ASS and recognise and maintain the fx's - I think that may be a tall order if not impossible. So you may have to recreate them in aegisub64 to create the ASS format subtitle to use or embed as a soft sub within a MKV file. Then it depends on your media player. Popcorn A-500 and the Zappiti 4K models do as standalones

They seem to be embedded in the MKV file. The image below is what you see in VLAN when you pick Subtitle Track 6 that's in the raw MKV rip. It's also exactly what you see if you play the blu-ray and use the menu to select Play Sing-a-Long version. I also need these to work using Plex.

What about KODI does it support image formats? Maybe the OP could try KODI instead of Plex apart from getting a suitable standalone media player with a video jukebox function?

The following formats are fully supported either as embedded tracks or external subtitle files. Full support means they are compatible with all Plex Apps, including clients that require transcoded media.
SRT (.srt)
SMI (.smi)
SSA (or ASS) (.ssa or .ass)
WebVTT (.vtt)
Other formats such as VOBSUB, PGS, etc. may work on some Plex apps but not all.

I did take the RAW MKV (which is 25GB so just way too big) and dump it into Plex. It lists the subtitles (all of them) as PGS. The 6th one works exactly as expected with one huge problem: Plex becomes very unstable. I needed to reboot after using it once and couldn't FF or RW without Plex going into a dead-end of just forever showing me the spinning (working) icon.
I'm also brand new here and based my post on another similar post I found here through google. It had no such MediaInfo report so I was unaware of that. Now I know:

Appearance has nothing to do with it.
PGS and Vobsub formats are a series of images not unlike thousands of jpg files. They can appear as any sort of image. SRT on the other hand are standard text just the same as you type an email. ASS are also a text format with added tags that allows for a range of effects very dependent on the media player. So I guess it is a problem with Plex and as they say not all Plex apps are equal.
PS If you want to reduce the size of the file a tad you could remove all the unnecessary tracks like languages and subs.
