Have you checked the 'Volume Mixer'? It may not have the audio set up for DVDs. Does it play a audio CD with the same drive? Vista has settings for each device, including the DVD/CD player.
If you go to the 'Control Panel>Sound' you should find the settings. (Classic View) Or if you have the icon on your Taskbar; R click on that.
What program are you using for playback? If you have Vista Premium or higher, it has the included DVD player. Otherwise, try VLC media player if Windows Media Player is being uncooperative. If VLC works, then you may have a codec problem.
EDIT: CDs use WAV audio. DVDs most often use Dolby AC3 audio. It may be you are missing the AC3 audio codec. Try installing AC3 Filter.
