Can anyone explain what the audio will be affected by selecting the Dolby Digital 5.1-ch, 2-ch or DTS-ch when setting the video compression of backup a DVD-9 to a DVD-5?

If you mean what I think you mean, you select what audio track(s) to keep. I usually select DD (AC3) 5.1 only.

Hey, is it necessary to check both of the Dolby Digital and DTS 5.1 when setting the Audio? If only choosing the DD 5.1, what will the audio lose?

It depends on the movie. Sometimes the 2.0 is the commentary tracks, sometimes it is the dolby prologic version of the audio. The audio does not get re-encoded or compressed, so select 5.1 means you will get the whole 5.1 audio, but nothing else.

You'll lose what you don't select. It will not be available on your final DVD. Most (all?) DVD players can downmix DD 5.1 to stereo if you don't have a 5.1 audio system hooked up to your player, so, if the DD track contains the audio you want, go for that. Like guns1inger says, any audio track can contain anything, so to be really sure, listen to it before deciding which to keep. (Even if I've never heard anyting but the "real" movie audio in the DD track.)
