I have a 3 hour TV program that will end up being about 2 1/2 hours.
The first VIDEO_TS folder will contain about 1hr40min and the second VIDEO_TS folder will contain about 50min.
Can I use DVD Shrink to combine into one VIDEO_TS?
If so, how?

So, in practice, you have 2 DVDs you want to author as one. Can be done with DVDShrink, Reauthor mode.

Then, you could:
Extract audio and video from all vobs in folder 1
Extract audio and video from all vobs in folder 2
(I'd use ReJig for this)
Author with TMPGEnc DVD Author (not freeware, I'm afraid...) with the A/V from the two folders as part of the same title. That way, it will play as one single movie.

It may give you more chapter markers than you want since it makes chapters based on cells. I've used it myself and it is very easy. Other than the extra chapters you might have...it is a great way to join two VIDEO_TS folders.
TMPGEnc DVD Author is a 30 day trial if I remember correctly. You may be able to remove the extra chapters with it. -
