Ok, say I have taken a laserdisc, captured in Huffyuv, converted to Mpg in Tmpgenc at 4800kb/s using 2-pass VBR and burned to a dvdr..
Now what if I was to rip that same dvd and run it through tmpgenc again at the same bitrate and 2-pass VBR--Would there be any quality gains?

Absolutley not. Everytime you re-encode (with a few exceptiosn for noise filtering) you lose quality.
You have the original Huffyuv capture, always work with that. If you need to improve the quality, try frameserving from VDUB and apply some filters. There's a number of things you can do to improve a laser disk capture quality. I was wondering if a another time through the encoder would smooth any spots it missed the first time--kind of silly I know but in this game, one never knows

vbr is good only for saving space. qulity will never be better than cbr with bitrate equal to max bitrate of vbr.
