Is it possible to run to instances of dvd2one at the same time?

Won't be quicker. The limiting factor is either the CPU, in which case running 2 at once will be slightly slower than 2 consecutively... or it's the hard drive, in which case running 2 at once will be MUCH slower than running them consecutively.
So the answer is, basically, don't do that. -
