    The source file is 640x272, 23.976 fps (widescreen)

    I am using virtualdub, frameserving, tmgenc 2.2 for conversion.

    Which source aspect ratio should I select in TMPGENC?
    When using 1:1 VGA, the sites (left and right) are cut of
    Using 16/9 PAL seems no good to me too?
    Does it matter that the frame rate of the mpeg (25 fps) isn't the same as the frame rate of the AVI?

    Should I select interlace or non interlace (in de video and advanced tab)

    Since you frameserve with, you should resize with Vdub as well. If you don't need any filters, there is no need to frameserve with Vdub.
    Resize to 480x320 and expand frame (add borders) to 480x576.
    TMPG> advanced> source aspect ratio 1:1 > video arrange method "full screen[keep aspect ratio]"
    Target res 480x576, aspect ratio 4:3.
    Since you convert the framerate, TMPG will duplicate ~1 fps, that may result in a jerky playback. My suggestion is, never convert the framerate unless you have no other choise.

    If you have to do this by all means I rather suggest to speed up the video a little. Unfortunately I don't know a VirtualDub filter, that will do this task. So you may try Avisynth. The complete script will look like this:

    AVISource("c:\filename.avi")
    BilinearResize(448,320)
    AddBorders(16,128,16,128)#put borders to TV overscan area
    AssumeFPS(25,sync_audio=true)#does not work with a seperate wav source
