Depends what is in the mkv. mkv files can contain numerous audio and video streams.

To play an mkv file you need
    1) an mkv parser filter such as Haali's found in the most recent mkv pack. VLC has one built in.
    2) A decoder for whatever video stream. VLC has numerous built in, however it can't handle everything that can be used in mkv.
    3) A decoder for the audio stream(s). VLC also includes various audio decoders.
    4) A subtitle filter if it uses subtitles (included in VLC).

Like I said it all depends on what is in the mkv file. There is no one codec that can decode all possible combos. mplayer can play I would think every mkv file though, including those containing Real Video. First player to support menus in mkv is I believe going to be VLC.
