How would one convert an AVI/ DV AVI from, say, Premiere to a usable format for plugging in to DVD2ONE? It's such a great program I would like to utilize it to create DVD's from my many hours of home videos.

Well my guess is that you can't do this. It seems like it's all based on DVD-ripped, and you can't do stuff like authoring with it. I believe you want the quick encoding, but I don't know if it's possible to reencode MPEG4 to MPEG2 like this. Guess you have to use another tool.
