Presumably you're referring to the "Basic Multiplex" and "Basic Demultiplex" settings. (I don't really expect anyone to copy it character-by-character...)
The Basic versions simplify the most common mux/demux task: multiplexing or demultiplexing a single video stream and a single audio stream. For such tasks, there's no benefit to using the more general Multiplex or Demultiplex tabs.
If you want to do something more sophisticated, however, like multiplex together a video stream and TWO audio streams, you'd use the general Multiplex tab. It lets you multiplex any combination of streams. I'll bet this would be helpful to someone making a DVD, who might want to multiplex multiple video streams, audio streams, and overlays for multiple camera angles, languages, and subtitles.
