I'm adding a 160 Gig drive just for video use. vcd- dvd- dv movie transfer etc.
Do I need to partition the second drive? Not sure how to procede. Thanks

Nope
You can leave it as one giant drive. I would break it up though. I have a 120gig drive with 3 partitions, PICTURES, MP3's/music, and one for DVD ripping work space. -
The new drive will have to contain some sort of partition, otherwise you will not be able to format it. Whether it is 1 big partition or 2 smaller ones is up to you. I would recommend you partition it as 1 drive partition and format using NTFS. If you try to partition and format as FAT32 w/ XP, you will be limited to a 32GB partition. NTFS does not have this limitation.
