I have an edited DV project with transitions etc. in Adobe Premiere Pro, ready for exporting to MPEG2.
Is there a quality difference if I directly export the timeline to a MPEG2 file or when I first export the timeline into a DV file and next export this file into MPEG2?

There would probably not be any noticeable difference in exporting the timeline back out to a DV AVI file and then encoding that DV file to MPEG2, however, the DV file is a compressed video file, so for the best quality, it would be best to go straight to MPEG2 from the timeline.
If you are considering using a different MPEG2 encoder as opposed to the Main Concept encoder (which I presume is still bundled with Premiere Pro), there are options to "frame serve" the timeline to the external encoder such as TMPGEnc, so that you don't have to create the intermediate DV AVI file. This website should have links to frameservers under the Tools section.
