I have an avi file that contains subtitles. I converted the avi file to mpeg2 and it plays perfectly on my computer with subtitles. Now I want to run the file through tmpgenc dvd author to create dvd compliant files, but I read, in a guide, that tmpgenc dvd author does not support subtitles. If I run it through tmpgenc dvd will I lose the subtitles?

No. They are part of the video, so they wont be affected. TMPGEnc DVD Author doesn't support DVD subtitles. They are stored in a separate stream in the VOB, and not at all retated to subs that are part of the AVI.
