  1. How do I use scripting in Avidemux and VirtualDub2? Can I use it with the GUI or not? What should the file format be for my scripts for Virtualdub?

  2. I don't use AviDemux but use AviSynth scripts in VDub every day. You create a script and open it as you would any video. File->Open Video file. Avisynth scripts use .avs as the suffix.
    Ordinarily, you don't use it with VDub's filters. All the filtering is best done within the script.

  4. That is a batch file (fragment), not a AviSynth or VirtualDub script. You can use that type of batch file with any program that accepts arguments on the command line.
