Is it possible to do this with an .srt or .ssa subtitle file? Was hoping I could get the more curly looking quotes instead of the straight ones.

Yes, open your srt in Notepad or whatever text editor you use. Global search and replace may be awkward though as straight quotation marks are the same whereas curly quotation marks are opening and closing. Whether your chosen video player will display them correctly, I've no idea.
Here are the Alt+ codes you need for curly quotes, use numeric keypad and note that NumLock key must be activated.
