  1. I'm editing a project in Adobe Premiere that I want to ultimately use DVD-Lab to author and then burn to disk. My question is I'm editing in Premiere in AVI, do I encode to MPEG-2 before authoring in DVD-Lab or after? Thanks, I'm new to this.

DVD-Lab is an authoring tool, so it needs already-encoded MPEGs which it demuxes. So YES, you need to encode to MPEG-2 before you author.
    If in doubt, Google it.

  3. it also takes elementary streams, just in case your mpeg2 video and your audio are separate

if they're separate they are called elementary streams, if both are contained in one file it is a program stream
