Your question is like asking a mechanic why your car won't start without him being able to look at it. It could be anything.

Copy the contents of MeGUI's log file and add it to a post. It's fairly informative and will usually provide some clues.
It's probably easiest to restart MeGUI, run a job that you know will fail, close MeGUI after it fails, then copy the entire contents of the newest log file in the MeGUI/logs folder.
