Whenever I try to play the ISO it looks like it works, but then immediately exits.
Do I have to update my MEncoder/MPlayer in some form?

Please post the exact message(s) that Mplayer displays before exiting.
Example:

He's right. Need the log file.
Are you actually using mplayer from the command line or using a front end for it? The reason I'm asking is that if you're using it in command line mode I'd think you'd already have figured out searching "mplayer play iso" and "mplayer log file". Most mplayer front ends like smplayer will have the option to save/display mplayer crash logs.
iso playback has been supported in mplayer for at least 5 years. You shouldn't need an update.
