I joined a movie site and on the movie there is choice to download the clip as a AVI 320p. You can watch the movie in flash in 480p. I have Video DownloadHelper and can download the flash file in 480p but would it be better since its 420p? The AVI file is bigger though don't know if that makes a difference though.

Download both and see which looks better. There's no way of saying for sure otherwise.
A larger file size SHOULD indicate more bitrate was used and thus should be of higher quailty. However a codec was not mentioned. Also the source isn't mentioned. If its not a clean source it won't really matter what file you choose.
But all things being equal a file at a lower bitrate and the same codec will not look as good as a file at a higher bitrate and the same codec (and the same resolution of course).
And jagabo is correct you will have to determine this yourself. Just do a comparison test.Donatello - The Shredder? Michelangelo - Maybe all that hardware is for making coleslaw?
And note the importance of "all (other) things being equal". Different codecs have different abilities to compress video. Even the same codec can deliver different quality depending on its settings. If the encoders used different sources (say VHS, vs Blu-ray) there could be big difference even with the same codec and the same settings.
Which is faster, a Volkswagon or a Ferrari? Obvious huh? What if the Ferrari has no wheels? What if its engine is blown. What if its upside down, standing on its roof? You see, there are a lot of details that need to be considered.
