Basically I'm asking if there is one tool that will do it without going through intermediate steps or processing? What I'm doing now is converting an 8GB MKV file to DVD9 size using VSO ConvertXtoDVD and then using DVDShrink to make an uncompressed ISO.

Probably not. Check what your mkv consists of with mediainfo
Assuming you mean ISO of a DVD-9 that uses DVD-Video (i.e. standard def. and playable in a DVD player), your mkv would have to be MPEG2 based. (It probably isn't, and you would have to convert h.264=>MPEG2 and then author to DVD-Video, and then wrap into ISO)
If you meant an ISO image of a DVD-9 that uses AVCHD (HD and playable in blu-ray player), then you could do it in fewer steps, because your wouldn't have to convert h.264=>MPEG2 -
I play everything over my network to my Beyonwiz PVR P1. Unfortunately at the moment the Beyonwiz http://www.beyonwiz.com.au/BWhomepage.html doesn't handle very well the wide range of MKV files that exist. The next after next firmware update should address that problem. It does however handle ISO and DivX and MP4 and MPEG PS and MPEG TS.

Can it handle h.264 transport stream? If so , just re-wrap the mkv (I'm going to assume it's using h.264) into MPEG TS (like .m2ts or .ts) with tsmuxer. This should take a few minutes instead of over an hour doing the conversion
An ISO is just a disc image. It can be an ISO of a blu-ray BD50 or BD25 for example. The term "ISO" isn't necessarily restricted to DVD5/9 and not video either (e.g you can have an ISO of a video game DVD, or a CD image too) . You probably mean DVD-Video ISO in this context for your player. -
Iso is more than just a disc image,its a file container that can hold any number of files with the idea of just one file being used for purposes such as streaming and mounting,if you put the mkv in iso form its up to the player or device to read it if it can.
Some read dvd iso as discussed,being just a single file it doesn't make much sense putting a single mkv into iso but a few mkv would be good if any reader could read it.I think,therefore i am a hamster. -
Thanks I will give it a go - however my i7 + Windows 7 is very fast. VSO ConvertXtoDVD takes all of 18 minutes and DVDShrink a further 10 minutes. So make a cup of coffee and voila it's done!
