Can anyone recommend an application to organize and filter a large library of videos? Basically I want to organize quicktime files of commercial spots for a TV network. I'm thinking something with the functionality of iTunes but on a professional level. It'd also be great if I had the ability to enter in my own set of metadata. We're talking 5,000 spots or so, average length 50 seconds.

You could always custom-build an MS Access Database. Then, you can fairly easily modify as your needs (fields) change.
(Have to know how to create a well-designed database, but it's not that hard)
