VLC plays most of the MKV files that I've come across.

Quicktime (with Matroska and Perian components) and VLC are able to read most mkv files, providing they do not include any RealMedia track (RV40...). VLC is better, though, because you can easily choose your language in case of multiple audio/subtitle tracks.

If the file contains RealMedia tracks (which is most probably the case) you'll have to use mplayerOSX with the Helix Real libraries. I don't know if this works for Intel based Macs.

If you want to do some conversion, use MKVExtract, then process the tracks with ffmpegX.
