There is a rule that we should substract Audio Size from Total size to find out Final Size of a Video File on CD after convertion is done. How can I find out how large is Audio Size of a video file when I convert the video file by Virtual Dub (two pass converting), because I want to know the final size of the file on CD?

If the audio is already embedded within the file...then you already know the final size.

Also the size is more than just audio + video. There is also container overhead.

Audio streams are either LPCM, CBR compressed or VBR compressed.
If they're VBR, you won't know until you can figure out the total length and the AVERAGE bitrate.
If they're CBR or LPCM, it's easy:
Filesize = Length of time x Bitrate
So, for example, 20.5 minutes of Audio CD LPCM data (headerless WAV type) is ~1378kbps * 1230 sec = ~206MB.
-or-
98 minutes of 384kbps CBR-compressed 5.1/AC3 is 384kbps * 5880sec. = ~276MB.
