Encode video to DVD specs mpg.
    Load it in DVD-Lab Pro.
Load the .srt in DVD-Lab Pro.
    Author.

    Or:
    Substitute DVD-Lab Pro for DVDAuthorGUI.

    Or: (for permanent subs)
    Load AVI in VirtualDub
Add the .srt using the TextSub filter
Frameserve to encoder and encode to DVD specs mpg.
    Load mpg in DVD-Lab
    Author

    Or:
    Use AviSynth instead of VirtualDub.

mats.hogberg gave a few excellant options, but if you must do your layout in DVDLab, convert your .srt subs to .sup, create a separate DVD layout using something like muxman that includes the subs, then use Vobblanker to replace the title using the version with subs but sticking with your menu and such from DVDLab. There's guides here on specifics.
