I have one big VOB file (2,5 GB) which I would like to burn on a DVDR.
Nero syas that the VOB file is too big. What can I do??

The problem (and reason that Nero says it is too big) is that VOB files are a total of 1GB each; I think this is to avoid compatibility problems with legacy systems. (The ISO standard, BTW, only supports files of up to 2GB each).
How you ended up with a 2.5GB vob, I don't know, but try running it through DVD Shrink at 100 or 99% and see if it produces a correct series of VOBs.
