I ripped a chapter from a DVD using DVD Shrink. I then used DVD2AVI so that I could open the file in TMPEnc and compress to my choosing.
DVD2AVI provided two files:
i) test.d2v
ii) test AC3 T01 2_0ch 192Kbps DELAY -48ms.ac3
Since test.d2v is the video file and works as a "video source" in TMPGEnc I presume the other one is the audio file.
However, this does not seem to be a valid audio source since when I try it, I receive an error message stating it could not be opened or is unsupported.
Any ideas?
Or any other program I could use to covert/compress the ripped chapter?

Depends what you want on your eventual output file?
TmpgEnc cannot have an ac3 file as a source for encoding. However you could just encode the video and multiplex the ac3 back in afterwards.
If you want to encode the audio choose the option Decode in the audio menu in DVD2AVI instead of Demux. This will give you a .wav file instead of ac3 (but you will lose the 5.1 if you had it) which can be loaded into TmpgEnc -
On DVD2AVI there are settings to ouput WAV instead of AC3.
Before you save project, click on 'Audio / Output Method / Decode to Wave'.
The default setting will 'Demux' to AC3. -

TMPGEnc gives the "Cannot open, or unsupported" error when you don't have a codec installed. Try installing AC3Filter.
Are you re-encoding the file in TMPGEnc to make a VCD/SVCD/DVD ?
If DVD, you can leave the audio as-is, as it already is DVD-compliant - or if you want to reduce the bitrate, or if you're making VCD/SVCD then use ffmpeggui or besweet or belight to convert the file to MP2/AC3.If in doubt, Google it.
