If I want to make a 5.1 audio in Premiere PRO, how shall I do??

Yes you can make 5.1 audio, it is one of the new features in the latest version.
I haven't done it myself, but there is a plugin which you may have gotten with your premiere installation called SurCode which is an AC3 audio encoder. It is only a trial version which gives you 3 uses I think, check it out anyway.
