have just bought a second hand 500 u burner but its locked on region have used latest firmware and have tried all sorts of region killers but no joy

yep its probably locked onto that last region
not much you can do about it YET, but just grab a cheap dvd-rom drive like a liteon.
Ive had my dru500a for about 5 months and i havent even set the region on the drive yet. Why? Cause i dont really need to. Its ripping speed of dvd is quite crap at 2x dvd-video or 4x dvd-r, where as the liteon can hit 16x on single layer disc, or 8x on dual layer disc, as well as climb into the 10x region on dvd-r discs... MUCH MUCH faster and no wear on the expensive dvd writer. And the dvdrom drive is region free. Win Win if you ask me
