    Is this possible?

Is it as simple as changing a file extension?

No! A DVD consists of many files BUP, IFO and VOB's. What you can do is put a DVD format into a container like MKV, no reencoding or loss of quality but it will be the same size. It is then a "single file."
    An example of a "DVD"

The Man From Snowy River DVD
    AUDIO_TS (Folder)
VIDEO_TS (Folder containing the following files)
    VIDEO_TS.BUP
    VIDEO_TS.IFO
    VTS_01_0.BUP
    VTS_01_0.IFO
    VTS_01_1.VOB
    VTS_01_2.VOB
    VTS_01_3.VOB
    VTS_01_4.VOB
    VTS_01_5.VOB

You could alternatively select the VOB's relating to the main feature and use a program like VOB2MPG to turn them into a mpg file. The easiest IMO is to use MakeMKV and select the main feature.
