I usually use VirtualDub to get screenshots from a video for every 500 frames but virtualdub can't open mkv files. Is there any other program that can open mkv files and take screenshots of every 500 frames?

Try virtualdubmod or try Video Thumbnails Maker.
I can't see an option to set how many frames to screen capture in video thumbnails maker
I've always used MPC. But I don't know if it's adjustable.

MPC only does thumbnails. I wanted full size screenshots of every 500 frames
You could probably do it with AviSynth using SelectEvery() and ImageWriter().

You can also open it up with an avisynth script in virtualdub. You need avisynth & the proper splitters & decoders installed (e.g. haali media splitter, ffdshow). Just write the text file, rename extension to .avs. Open the .avs in virtualdub

VLC does a good job on screen grabs as well. It also helps smooth interlaced frames so the image doesn't suffer from interlaced artifacts.
